Preparation of Synaptosomes and Synaptic Vesicles from Rat Brain

ثبت نشده
چکیده

Isolated nerve terminals (synaptosomes) were prepared from adult and embryonic Wistar rat whole brains or isolated brain areas following the procedure described by Edelmann et al. (1995). Mostly, pregnant female rats and their embryos were used, although synaptosomes were also occasionally prepared from male rats. Usually, one rat was sufficient for a synaptosomal preparation. The rat was anaesthetised with ether and immediately sacrificed by decapitation. The adult brain was subsequently removed as quickly as possible and instantly placed in ice-cold sucrose solution (see Section 2.3). Pregnant females generally carried between 15 and 18 embryoes, which were removed and placed on foil-covered ice. Embryonic brains were isolated as fast as possible and pooled in ice-cold sucrose solution. All subsequent steps were then performed on ice or at 4 °C.

Two distinct populations of synaptic-like vesicles from rat brain.

In nonneuronal cells, several plasma membrane proteins such as exofacial enzymes, receptors, and ion channels recycle between their intracellular compartment(s) and the cell surface via an endosomal pathway. In neurons, however, this pathway has not been extensively characterized. In particular, it remains unclear whether or not it is related to the recycling of small synaptic vesicles, the maj...

The effects of JM-20 on the glutamatergic system in synaptic vesicles, synaptosomes and neural cells cultured from rat brain

JM-20 (3-ethoxycarbonyl-2-methyl-4-(2-nitrophenyl)-4,11-dihydro-1H-pyrido[2,3-b][1,5]benzodiazepine) is a novel benzodiazepine dihydropyridine hybrid molecule, which has been shown to be a neuroprotective agent in brain disorders involving glutamate receptors. However, the effect of JM-20 on the functionality of the glutamatergic system has not been investigated. In this study, by using differe...

Synaptophysin binds to physophilin, a putative synaptic plasma membrane protein

We have developed procedures for detecting synaptic vesicle-binding proteins by using glutaraldehyde-fixed or native vesicle fractions as absorbent matrices. Both adsorbents identify a prominent synaptic vesicle-binding protein of 36 kD in rat brain synaptosomes and mouse brain primary cultures. The binding of this protein to synaptic vesicles is competed by synaptophysin, a major integral memb...

High and low temperatures affect rat hippocampal synaptosome’s viability and functions

Introduction: Synaptosomes are sealed particles that contain mitochondria, cytoskeleton and vesicles which are necessary to synaptic events like neurotransmitter release and uptake in the nervous system. However, the effect of high and low temperatures on synaptosome membrane integrity and function during a time course after its extraction is less known. The purpose of this study was to assess ...

Effect of Paraoxon on GABA Uptake by Rat Cerebral Cortex Synaptosomes

Background: It has been suggested that organophosphates may inhibit gamma-aminobutyric acid (GABA) metabolism in synaptosomal preparations. In the present investigation, we have assessed the interaction between paraoxon and the GABA system at synaptic level. Methods: Synaptosomes were prepared from male Wistar rats (200-250 g). Cerebral cortex was dissected and homogenized, then centrifuged at ...
